Polyethylene-Oxide (PEO) Linear Nano-Polymer Directly Stimulates Endothelium and Protects Myocardium from Focal Ischemia-Reperfusion Injury via eNOS Pathway in Rats

چکیده
Medical grade Nano-polymers (NP) used to enhance circulation, drug and gene delivery. Injection of Polyethylene-oxide (PEO) increase tissue perfusion and decrease mean blood pressure in animal models. Polyethylene-oxide (PEO) NP’s ultra high molecular weight (2-5 x 10^6 Da) and rheological properties allow it to interact with endothelial cells to increase endothelial nitric oxide (eNO) and its attendant enzyme (eNOS). eNOS over-expression and eNO induction is known to provide cardioprotection. We hypothesized that injection of PEO will have cardioprotective properties in a model of myocardial ischemia reperfusion injury (I/R), via induction of eNOS due to direct stimulation of endothelium. Male rats (310±9 g, n=16) were randomized to IV injection of 10 ppm of a saline-PEO solution (PEO) or equal volume of saline (CONT). Focal I/R injury was induced by LAD ligation for 30 minutes followed by 60 min of reperfusion. ECG, and aortic pressure monitored continuously and infarct size calculated as % area at risk. eNOS and its active phosphorylated form (p-eNOS) were detected on Western blots in left ventricle protein extracts. Mean blood pressure was 78±10 and 75±6 in PEO compared to 58±12 and 66±4 in CONT, at 30 min of Ischemia and 60 min reperfusion respectively (p<0.05 PEO vs. CONT). PEO had 28% and 47% higher eNOS and p-eNOS vs. CONT. PEO had a 28% infarct sparing effect compared to CONT. PEO prior to myocardial infarction significantly reduces infarct size and induces activation and up regulation of eNOS pathway. This is a possible novel mechanism for PEO NP cardioprotective effect and behavior in blood circulation. These properties of PEO warrant further investigation.

Postconditioning: a form of "modified reperfusion" protects the myocardium by activating the phosphatidylinositol 3-kinase-Akt pathway.
Brief intermittent episodes of ischemia and reperfusion, at the onset of reperfusion after a prolonged period of ischemia, confer cardioprotection, a phenomenon termed "ischemic postconditioning" (Postcond). We hypothesized that this phenomenon may just represent a modified form of reperfusion that activates the reperfusion injury salvage kinase (RISK) pathway. Isolated perfused rat hearts were...
متن کاملDirect Renin Inhibition With Aliskiren Protects Against Myocardial Ischemia/Reperfusion Injury by Activating Nitric Oxide Synthase Signaling in Spontaneously Hypertensive Rats
BACKGROUND We tested the hypothesis that direct renin inhibition with aliskiren protects against myocardial ischemia/reperfusion (I/R) injury in spontaneously hypertensive rats (SHR), and examined the mechanism by which this occurs. METHODS AND RESULTS Male SHR were treated (orally, 4 weeks) with saline or aliskiren (30 or 60 mg kg(-1) day(-1)) and subjected to 30 minutes of left anterior des...
متن کاملNorth American ginseng protects the heart from ischemia and reperfusion injury via upregulation of endothelial nitric oxide synthase.
Emerging evidence suggests ginseng has therapeutic potential in cardiovascular disease. The aim of this study was to investigate the role of endothelial nitric oxide synthase (eNOS) in the cardioprotective effects of ginseng during myocardial ischemia and reperfusion (I/R). Treatment with ginseng extract significantly increased Akt phosphorylation and eNOS protein levels in cultured neonatal ca...
متن کاملBlockade of Central Angiotensin II AT1 Receptor Protects the Brain from Ischemia/Reperfusion Injury in Normotensive Rats
Background: Stroke is the third leading cause of invalidism and death in industrialized countries. There are conflicting reports about the effects of Angiotensin II on ischemia-reperfusion brain injuries and most data have come from chronic hypertensive rats. In this study, hypotensive and non-hypotensive doses of candesartan were used to investigate the effects of angiotensin II AT1 receptor b...
متن کاملPPAR-alpha activation protects the type 2 diabetic myocardium against ischemia-reperfusion injury: involvement of the PI3-Kinase/Akt and NO pathway.
Several clinical studies have shown the beneficial cardiovascular effects of fibrates in patients with diabetes and insulin resistance. The ligands of peroxisome proliferator-activated receptor-alpha (PPAR-alpha) reduce ischemia-reperfusion injury in nondiabetic animals. We hypothesized that the activation of PPAR-alpha would exert cardioprotection in type 2 diabetic Goto-Kakizaki (GK) rats, in...
